Establishment and in vitro germination of Churuja orange pitahaya (Hylocereus sp.) seeds

The objective of this research was to establish and germinate in vitro seeds of Churuja orange pitahaya (Hylocereus sp.), allowing young plant material to be obtained under aseptic conditions. Pitahaya seeds were introduced in in vitro conditions, using 70% ethanol at different times (seconds) and continued disinfection in different disinfectant solutions (sodium hypochlorite concentrations) and in different time periods (minutes). A completely randomized design (CRD) was used, establishing five treatments with 10 experimental units or replicates per treatment; the experimental unit corresponded to a plate with 15 seeds. As a result, it was observed that the best treatment allowed obtaining 0% contamination and 100% seed germination. In conclusion, a protocol for the establishment and in vitro germination of Churuja orange pitahaya (Hylocereus sp.) seeds were established. Through the use of a 70% ethanol solution for 1 minute, 2% sodium hypochlorite solution for 10 minutes and three washes in sterile distilled water (Treatment T5), obtaining contamination-free seedlings (0%) and total germination (100%) of Churuja orange pitahaya seeds.
